About the Project Prostate cancer frequently metastasises to bone, causing considerable morbidity and mortality. Once in the bone, prostate cancer cells disrupt normal bone turnover leading to clinically osteosclerotic lesion with underlying osteolysis. Tumour cells exhibit altered post-translational modification of many proteins, which affect tumour cell behaviours including invasion and metastasis. Specifically, N-glycans are a group of post-translational modification that are altered in prostate cancers and affect bone metastasis. A key enzyme in the formation of mature N-glycosylations is Fucosyltransferase 8 (Fut8). Fut8 is overexpressed in prostate cancers and has been strongly linked to bone metastasis. The detection and imaging of N-glycans by mass spectrometry imaging is in its infancy and this cutting-edge technique has been recently established in Sheffield. We will use two models of prostate cancer grown in vivo and in advanced cell culture models. We aim to: Assess whether current methods of tissue processing of in vivo tissues affect N-glycan and metabolite detection by mass spectrometry imaging Compare glycomic and metabolomic landscape of cells grown in 2D cell culture vs 3D cell co-culture with bone marrow stromal cells vs in vivo bone metastasis, Assess the impact of N-glycosylation-modifying drugs (including Fut8 inhibitors) on N-glycans in vivo vs 3D cell culture and d) assess the impact of FUT8 knockdown on N-glycans in vivo vs 3D cell culture. Detection of glycans and metabolites will be performed by Desorption Electrospray Ionisation Mass spectrometry imaging (DESI-MSI) and/or Matrix-assisted Laser-desorption Ionisation Mass spectrometry imaging (MALDI-MSI). Glycan detection on tissues will be detected following Peptide N-glycosidase F (PNGase) or EndoF3 digestion of tissues prior to MSI detection of cleaved glycans. The glycomic and metabolomic landscape of in vivo tissues generated from a FUT8 knock-down PC3 cell line grown in bone, along with syngeneic prostate cancer cells grown in bone will be used and compared with a FUT8 inhibitor, along with comparisons of in vivo prostate cancer cells in bone with 3D cell culture of prostate cancer/bone marrow stromal cell co-cultures. A tax-free annual stipend at the standard UKRI rate (£19,237 for 2024/25 entry), research costs and tuition fees at UK rate Academic year: 2025/26 Open to: International (including EU) and UK (home) students The studentships are fully funded for four years, and you must complete your PhD in four years. You'll receive the UKRI minimum doctoral stipend per year for your living costs, which is paid to you in regular instalments.
